Biological Background: CCL19 Function and Localization
- CCL19, also known as ELC, MIP-3β, or CKβ11, is a secreted C-C motif chemokine involved in immune cell trafficking and inflammatory responses.
- It plays a critical role in normal lymphocyte recirculation and homing, particularly mediating T-cell and B-cell migration to secondary lymphoid organs via binding to chemokine receptor CCR7.
- Recombinant CCL19 exhibits potent chemotactic activity specifically for T-cells and B-cells, but not for granulocytes or monocytes.
- Additionally, CCL19 binds to atypical chemokine receptor ACKR4, triggering the recruitment of β-arrestin (ARRB1/2).
- The protein is highly expressed in lymph nodes, thymus, and appendix, with moderate levels in colon and trachea, and low levels in spleen, small intestine, lung, kidney, and stomach.
- CCL19 is secreted and contains disulfide bonds characteristic of chemokines, as indicated by keywords: Secreted, Signal, Disulfide bond.
Experimental Guidance and Technical Tips
- For western blotting (WB), note that the predicted molecular weight of CCL19 is approximately 11 kDa, but the mature secreted protein may migrate differently due to post-translational modifications; consider using a low-molecular-weight marker and optimizing gel percentage.
- In immunofluorescence on paraffin-embedded sections (IF-P), CCL19 expression is highest in lymphoid tissues; validate antibody specificity in positive control tissues such as lymph node or thymus.
- For ELISA applications, this antibody may be suitable as either capture or detection reagent; however, perform standard curve and matrix effect validation in the relevant sample system.
- The antibody cross-reacts with mouse and rat CCL19, enabling its use in animal models; confirm cross-reactivity in your specific application.
