Biological Background: CCL17 Function and Localization

  • C-C motif chemokine 17 (CCL17) is also known as thymus and activation-regulated chemokine (TARC), small-inducible cytokine A17, and is encoded by the CCL17 gene.
  • CCL17 displays chemotactic activity for T lymphocytes, preferentially Th2 cells, but not monocytes or granulocytes, playing an important role in inflammatory and immunological processes. Related references: PMID:8702936 PMID:9169480
  • It acts by binding to the CCR4 receptor at the T-cell surface. Related references: PMID:10540332 PMID:9169480
  • CCL17 mediates GM-CSF/CSF2-driven pain and inflammation. Related references: PMID:27525438
  • In the brain, CCL17 is required to maintain the typical, highly branched morphology of hippocampal microglia under homeostatic conditions and may be important for the appropriate adaptation of microglial morphology and synaptic plasticity to acute neuroinflammation.
  • CCL17 plays a role in wound healing, mainly by inducing fibroblast migration into the wound.
  • Constitutively expressed in the thymus, with lower levels detected in the lung, colon, and small intestine; expression is upregulated in stimulated peripheral blood mononuclear cells. Related references: PMID:8702936
  • The protein is secreted and contains characteristic disulfide bonds typical of chemokines.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 24-94 of human CCL17, covering the mature chemokine domain. For western blot, consider that the mature secreted form may be smaller than the predicted 11 kDa due to signal peptide cleavage; a band around 8–11 kDa could be expected depending on the sample and detection conditions.
  • CCL17 is a secreted cytokine; for WB or ELISA, analyze cell culture supernatants, tissue homogenates, or serum/plasma. Ensure appropriate sample preparation to avoid degradation.
  • Given the listed cross-reactivity with mouse and rat, validate the antibody’s performance in these species using positive controls or recombinant proteins.
  • For ELISA, the antibody can be used as a capture or detection reagent; optimal pairing with another CCL17-specific antibody or standard may be necessary.
  • Post-translational modifications and dimerization can influence migration in WB; consider using reducing and non-reducing conditions to confirm specificity.
