Biological Background: CCKBR Function and Localization
- CCKBR (Cholecystokinin B receptor) is a G protein-coupled receptor that serves as a receptor for the peptide hormones gastrin and cholecystokinin (CCK). It is also known as CCK2-R and is encoded by the CCKBR gene in humans.
- The receptor is widely expressed in the central nervous system where it modulates anxiety, analgesia, arousal, and neuroleptic activity.
- CCKBR couples to both GNAI1 and GNAQ signaling pathways, but not to GNAS (PubMed:34556863). Related references: PMID:34556863
- In the gastrointestinal system, gastrin-activated CCKBR reduces glucose absorption in intestinal epithelial cells by downregulating SGLT1 and GLUT2 expression through suppression of the PI3K/Akt/eIF4B pathway. In the kidney, it decreases SGLT2 expression under high-glucose conditions via ERK/NF-kappa-B signaling.
- Isoform 2 is constitutively activated and may regulate cancer cell proliferation through a gastrin-independent mechanism. Isoform 3 is expressed in brain, pancreas, stomach, and certain cancer cell lines.
- Post-translational modifications include glycosylation and palmitoylation, and the receptor contains disulfide bonds and transmembrane helices typical of GPCRs.
- Subcellular localization is primarily at the cell membrane. Tissue specificity: Isoform 1 is expressed in brain, pancreas, stomach, colon cancer cell line LoVo, T-lymphoblastoma Jurkat, and small cell lung cancer lines; not in heart, placenta, liver, lung, skeletal muscle, kidney, or stomach cancer AGS. Isoform 2 is found in pancreatic and colorectal cancer cells but not normal tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 240-335 of human CCKBR (NP_795344.1), a region that may include intracellular domains. As a polyclonal antibody, it is expected to recognize multiple epitopes, potentially offering robust detection in denaturing and native conditions.
- For Western blot (WB), the predicted molecular weight of CCKBR is approximately 48 kDa (UniProt: 48,419 Da). Consider using appropriate blocking agents and optimizing antibody dilution to minimize background.
- Cross-reactivity is reported for Human, Mouse, and Rat. Validate in the relevant sample system to confirm performance, as expression levels may vary across tissues and cell lines.
- For ELISA, the antibody's polyclonal nature may enhance signal amplification. Consider plate-coating strategies and detection antibody pairing for sandwich ELISA development.
- Store at recommended temperatures (ship on ice) and avoid repeated freeze-thaw cycles to maintain antibody integrity.
