Biological Background: CCAR1 Function and Localization
- CCAR1, also known as Cell cycle and apoptosis regulatory protein 1 (CARP-1) or Death inducer with SAP domain, is encoded by the CCAR1 gene. Multiple isoforms exist due to alternative splicing.
- Plays a dual role as a transcriptional coregulator, associating with Mediator and p160 coactivator complexes to transduce signals from upstream activators to basal transcription machinery.
- Functions as a coactivator for p53 and nuclear receptors, including AR, where it can act as both coactivator and corepressor. In prostate cancer cells, CCAR1 contributes to chromatin looping and AR transcription complex assembly, stabilizing AR-GATA2 association and facilitating MED1/RNA polymerase II recruitment. Related references: PMID:23887938
- Involved in apoptosis signaling induced by the retinoid CD437, sequestering 14-3-3 proteins and altering expression of cell cycle regulatory genes like MYC, CCNB1, and CDKN1A. Related references: PMID:12816952
- During erythroid differentiation, CCAR1 enhances GATA1- and MED1-mediated transcriptional activation from the gamma-globin promoter in association with CALCOCO1. Related references: PMID:24245781
- Localizes primarily to the perinuclear cytoplasm.
- Expressed in various epithelial cancer cell lines (breast, colon, prostate, pancreatic, leukemia) and its expression is regulated by growth factors.
- Keywords indicate CCAR1 is a phosphoprotein subject to isopeptide bond formation and ubiquitin-like conjugation, suggesting regulation by post-translational modifications.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-200 of human CCAR1 (NP_060707.2), an N-terminal region that may be conserved across splice variants. Consider sequence alignment to assess isoform coverage.
- For Western blot (WB), CCAR1 has a predicted molecular weight of ~133 kDa. Given potential post-translational modifications and isoform diversity, multiple bands or size shifts may be observed; validate with appropriate positive and negative controls.
- In immunoprecipitation (IP), the rabbit polyclonal format may capture CCAR1 and its interacting partners. Optimize antibody concentration and lysis buffer conditions for your target complex.
- ELISA application is specified; ensure proper coating and detection conditions are established in your assay system.
- Cross-reactivity with mouse and rat indicates potential utility for studies in these species; however, verify the antibody's reactivity in your specific experimental context using species-appropriate controls.
