Biological Background: CCAR1 Function and Localization

  • CCAR1 (Cell division cycle and apoptosis regulator protein 1), also known as CARP-1 or DIS, is a multi-functional protein involved in transcriptional regulation, apoptosis, and cell cycle progression.
  • It functions as a transcriptional coactivator and corepressor, associating with Mediator and p160 coactivator complexes to modulate nuclear receptor and p53-dependent transcription (By similarity).
  • CCAR1 induces apoptosis in the presence of retinoid CD437 by sequestering 14-3-3 proteins and altering expression of cell cycle regulators such as MYC, CCNB1, and CDKN1A, thereby influencing cell proliferation. Related references: PMID:12816952
  • During erythroid differentiation, CCAR1 cooperates with CALCOCO1 to enhance GATA1- and MED1-mediated transcriptional activation of the gamma-globin gene in K562 cells. Related references: PMID:24245781

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-200), which may allow detection of multiple CCAR1 isoforms generated by alternative splicing. Confirm specificity in the relevant cell line.
  • For Western blot (WB), CCAR1 has a predicted molecular weight of ~133 kDa. Use appropriate molecular weight markers and loading controls, and optimize blocking and antibody dilution.
  • For immunoprecipitation (IP), consider using a standard protocol with protein A/G beads; validate the antibody's IP efficiency in your experimental system.
  • For ELISA, use recombinant CCAR1 protein or cell lysates to optimize coating and detection conditions.
