Biological Background: CBX1/HP1 beta Function and Localization
- CBX1/HP1 beta, also known as heterochromatin protein 1 homolog beta (HP1β), p25beta, and modifier 1 protein (M31), is encoded by the CBX1 gene and belongs to the chromobox protein family.
- It is a key component of heterochromatin, where it specifically recognizes and binds histone H3 tails methylated at Lys-9 (H3K9me), thereby contributing to epigenetic repression. Related references: PMID:41094145
- CBX1/HP1 beta also recognizes and binds histone H1.4 methylated at Lys-26 (H1.4K26me). However, simultaneous phosphorylation at Ser-27 (H1.4S27Ph) excludes it from chromatin. Related references: PMID:16127177
- Interaction with lamin B receptor (LBR) can mediate the tethering of heterochromatin to the inner nuclear membrane (By similarity).
- Subcellular localization is predominantly nuclear, consistent with its role in chromatin organization.
- It is widely expressed in all adult and embryonic tissues.
- Post-translational modifications include phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation, highlighting its dynamic regulation.
Experimental Guidance and Technical Tips
- The immunogen for this antibody is a recombinant fusion protein covering amino acids 1-100 of human CBX1/HP1 beta. Consider verifying specificity by knockdown or overexpression in your experimental system.
- For Western blot (WB), a band at approximately 21 kDa is expected. Use appropriate positive controls (e.g., human, mouse, or rat tissue/cell lysates) and validate loading with housekeeping proteins.
- In immunohistochemistry (IHC-P), antigen retrieval may be necessary; optimize conditions for your specific tissue samples.
- For ELISA applications, titrate the antibody and include relevant standards; note that this antibody recognizes human, mouse, and rat CBX1.
- As a rabbit monoclonal antibody, it typically offers high specificity and lot-to-lot consistency.
