Biological Background: CBR1 Function and Localization
- NADPH-dependent reductase with broad substrate specificity, reducing carbonyl compounds such as quinones, prostaglandins, menadione, and various xenobiotics.
- Converts antitumor anthracyclines doxorubicin and daunorubicin to cardiotoxic metabolites doxorubicinol and daunorubicinol. Related references: PMID:15799708, PMID:17344335, PMID:17912391
- Can convert prostaglandin E to prostaglandin F2-alpha (by similarity).
- Binds glutathione, showing higher affinity for glutathione-conjugated substrates; catalyzes reduction of S-nitrosoglutathione. Related references: PMID:17344335, PMID:18826943

- The immunogen corresponds to a central region (aa 100–200) of human CBR1. The antibody may recognize epitopes within this region across species, consistent with known cross-reactivity to mouse and rat.
- For Western blotting, a band around 30 kDa is expected. Validate with appropriate positive (e.g., kidney lysate) and negative controls.
- In IHC-P, optimize antigen retrieval and antibody dilution for the tissue of interest; note that CBR1 is expressed in kidney at the protein level.
- For ELISA, the recombinant immunogen can serve as a positive control; ensure optimal coating conditions and consider potential interference from reducing agents due to the enzyme's affinity for glutathione.
