Biological Background: CBR1 Function and Localization
- Carbonyl reductase 1 (CBR1), also designated 15-hydroxyprostaglandin dehydrogenase, 20-beta-hydroxysteroid dehydrogenase, and PG-9-KR, is a member of the short-chain dehydrogenase/reductase family.
- Functions as an NADPH-dependent oxidoreductase with broad substrate specificity, reducing a variety of carbonyl compounds including quinones, prostaglandins, and xenobiotics.
- Mediates the reduction of antitumor anthracyclines doxorubicin and daunorubicin to the cardiotoxic metabolites doxorubicinol and daunorubicinol. Related references: PMID:15799708 PMID:17344335 PMID:17912391
- Can bind glutathione, enhancing affinity for glutathione-conjugated substrates, and catalyzes the reduction of S-nitrosoglutathione. Related references: PMID:17344335 PMID:18826943
- Participates in glucocorticoid metabolism, reducing cortisol and corticosterone to 20beta-dihydrocortisol and 20beta-corticosterone, which are weak NR3C1/NR3C2 agonists in adipose tissue. Related references: PMID:28878267

- Subject to acetylation, alternative splicing, and phosphorylation; involved in lipid metabolism.

- The immunogen was designed against the C-terminal region (aa 178–277) of human CBR1, which may confer high specificity. Consider sequence alignment with homologous proteins if cross-reactivity is a concern.
- For Western blot, the predicted molecular weight is approximately 30 kDa. Validate detection in your sample system with appropriate positive and negative controls.
- In IHC-P, antigen retrieval conditions may require optimization. Pilot experiments with known positive tissues (e.g., kidney) are recommended.
- ELISA applications may benefit from testing a range of coating antigen concentrations and antibody dilutions to achieve optimal signal-to-noise ratio.
