Biological Background: CBP80 Function and Localization
- Nuclear cap-binding protein subunit 1 (NCBP1), also known as CBP80 or 80 kDa nuclear cap-binding protein, is the large subunit of the cap-binding complex (CBC).
- The CBC binds cotranscriptionally to the 5' cap of pre-mRNAs and participates in multiple RNA processing events, including pre-mRNA splicing, translation regulation, nonsense-mediated mRNA decay (NMD), microRNA (miRNA) biogenesis, and mRNA export.
- CBP80 does not directly bind the capped RNA; instead, it stabilizes the cap-binding loop of NCBP2/CBP20, locking the CBC into a high-affinity cap-binding state. Additionally, CBP80 can associate with NCBP3 to form an alternative CBC that specifically mediates mRNA export, particularly during cellular stress.
- The CBC complex is essential for the pioneer round of translation, which is tightly linked to NMD. It enhances NMD through interaction with UPF1 and supports both exon-junction complex (EJC)-dependent and failsafe NMD pathways.
- CBP80 is involved in miRNA-mediated gene silencing and pri-miRNA processing through its interaction with SRRT/ARS2.
- The protein localizes to both the nucleus and cytoplasm, consistent with its roles in nuclear mRNA processing and export as well as cytoplasmic translation surveillance.

- Post-translationally modified by acetylation, phosphorylation, and ISGylation (isopeptide bond).

- The immunogen corresponds to the N-terminal region (aa 1-100) of human CBP80; homology with mouse in this region should be considered for cross-reactivity.
- For Western blot, the predicted molecular weight is approximately 92 kDa; validate band size in your sample system.
- For IHC-P, consider antigen retrieval conditions and appropriate controls.
- As a polyclonal antibody, evaluate lot-to-lot consistency for critical applications.
