Biological Background: CBLB Function and Localization

  • CBLB (Casitas B-lineage lymphoma proto-oncogene b) encodes an E3 ubiquitin-protein ligase that accepts ubiquitin from E2 enzymes and transfers it to substrate proteins, generally targeting them for proteasomal degradation. Related references: PMID:20525694, PMID:40101708
  • It is a critical negative regulator of T-cell receptor (TCR), B-cell receptor (BCR), and high-affinity immunoglobulin epsilon receptor (FCER1) signal transduction pathways, contributing to immune tolerance and anergy. Related references: PMID:40101708
  • In naive T-cells, CBLB inhibits VAV1 activation upon TCR engagement, imposing a requirement for CD28 costimulation for proliferation and IL-2 production (By similarity).   • In activated T-cells, CBLB inhibits PLCG1 activation and calcium mobilization upon restimulation, promoting T-cell anergy (By similarity).   • CBLB localizes predominantly to the cytoplasm. It is expressed in placenta, heart, lung, kidney, spleen, ovary, testis, fetal brain, liver, and hematopoietic cell lines, but not in adult brain, liver, pancreas, salivary gland, or skeletal muscle. Present in lymphocytes at the protein level.
  • Alternative splicing produces multiple isoforms, and the protein is subject to phosphorylation and metal-ion binding. It contains zinc-finger domains, a RING finger, and is classified as a transferase in the ubiquitin conjugation pathway.

  • The immunogen for this antibody is a synthetic peptide spanning amino acids 883–982 of human CBLB (isoform NP_001308717.1), a region near the C-terminus. Given the target's cytoplasmic localization, consider using appropriate lysis buffers and validating detection in whole-cell lysates.
  • For Western blot (WB), the predicted molecular weight of CBLB is approximately 109 kDa; inclusion of a prestained molecular weight marker is recommended. Since cross-reactivity is reported for human, mouse, and rat, anticipate a band at a similar size across these species, but always validate with positive and negative controls.
