Biological Background: CBLB Function and Localization

• CBLB (Casitas B-lineage lymphoma proto-oncogene b) encodes an E3 ubiquitin-protein ligase that accepts ubiquitin from E2 enzymes and transfers it to substrate proteins, generally targeting them for proteasomal degradation. Related references: PMID:20525694, PMID:40101708
• It is a critical negative regulator of T-cell receptor (TCR), B-cell receptor (BCR), and high-affinity immunoglobulin epsilon receptor (FCER1) signal transduction pathways, contributing to immune tolerance and anergy. Related references: PMID:40101708
• In naive T-cells, CBLB inhibits VAV1 activation upon TCR engagement, imposing a requirement for CD28 costimulation for proliferation and IL-2 production (By similarity). • For Western blot (WB), the predicted molecular weight of CBLB is approximately 109 kDa; inclusion of a prestained molecular weight marker is recommended. Since cross-reactivity is reported for human, mouse, and rat, anticipate a band at a similar size across these species, but always validate with positive and negative controls.
• In ELISA applications, the polyclonal nature of this antibody may capture multiple epitopes, enhancing sensitivity; however, titration and optimization of the coating and detection conditions are advised.
