Biological Background: CBL Function and Localization
- CBL encodes the E3 ubiquitin-protein ligase CBL, also known as Casitas B-lineage lymphoma proto-oncogene, c-Cbl, or RING finger protein 55. It is a proto-oncogene and member of the CBL family of RING-type E3 ubiquitin transferases.
- Functions as a negative regulator of numerous signaling pathways by catalyzing the ubiquitination and subsequent proteasomal degradation of activated cell surface receptors, including EGFR, KIT, PDGFRA, and PDGFRB. Related references: PMID:10514377, PMID:11896602, PMID:14661060, PMID:15190072, PMID:21596750.
- Recognizes and mediates ubiquitination of membrane-bound SRC family kinases, such as HCK and SRC, leading to their degradation. Related references: PMID:11896602.
- Plays an essential role in osteoblast differentiation and apoptosis, and is critical for osteoclastic bone resorption via a signaling pathway involving phosphatidylinositol 3-kinase activation. Related references: PMID:14739300, PMID:15190072, PMID:18374639.
- Participates in signal transduction in hematopoietic cells; also involved in LAG3-mediated T-cell regulation and HSV-1-induced NECTIN1 downregulation. Related references: PMID:28381567, PMID:40101708.
- Subcellular localization includes cytoplasm, cell membrane, cell projections (cilia), and Golgi apparatus. Post-translational modifications include phosphorylation (e.g., at Tyr-731) and autoubiquitination, which modulate its activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 850–906 near the C-terminus of human CBL; consider this region when designing detection strategies for full-length vs. truncated isoforms.
- Recommended for Western blotting (WB) due to the high molecular weight of CBL (~100 kDa); ensure adequate transfer conditions for large proteins.
- For immunohistochemistry (IHC-P), validate with appropriate positive and negative control tissues, considering CBL’s cytoplasmic and membrane localization.
- In immunoprecipitation (IP), confirm efficient capture of the target and co-immunoprecipitated partners under native conditions; the E3 ligase activity may be context-dependent.
- For ELISA, optimize coating and detection conditions using recombinant CBL protein as standard.
- Cross-reactivity with mouse and rat allows use in rodent model studies; but confirm sequence homology in the epitope region for reliable detection.
