Biological Background: CBL Function and Localization
- CBL (also known as c-Cbl, RNF55, CBL2) is an E3 ubiquitin-protein ligase and a proto-oncogene that functions as a negative regulator of multiple signaling pathways by mediating ubiquitination of cell surface receptors, leading to their proteasomal degradation. - Recognizes and ubiquitinates activated receptor tyrosine kinases (RTKs) such as KIT, FLT1, FGFR1/2, PDGFRA/B, CSF1R, EPHA8, and KDR to terminate their signaling. Related references: PMID:15190072 PMID:18374639 PMID:21596750

Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 850-906) of human CBL. For Western blot, expect a band at ~100 kDa; post-translational modifications may cause slight shifts.
- For immunohistochemistry (IHC-P), optimize antigen retrieval and antibody dilution experimentally; the antibody detects both membrane and cytoplasmic CBL.
- In immunoprecipitation, the polyclonal nature may offer robust pull-down efficiency; validate in the appropriate cell or tissue lysate.
- For IF/ICC, fixation and permeabilization conditions should be titrated to preserve both membrane and intracellular pools of CBL.
- Cross-reactivity with mouse and rat is reported; detection in other species may be possible but should be verified by sequence homology.
- As with any polyclonal antibody, lot-to-lot variability is possible; we recommend including appropriate controls and validating in the relevant sample system.
