Biological Background: CBFB Function and Localization
- Encoded by the CBFB gene, core-binding factor subunit beta is a transcription factor subunit that forms a heterodimeric complex with RUNX family proteins (RUNX1, RUNX2, RUNX3) known as core-binding factor (CBF). CBFB allosterically enhances the DNA-binding affinity of RUNX transcription factors.
- CBF complexes bind to the core consensus sequence 5′-TGTGGT-3′ in enhancers and promoters of various genes, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, LCK, IL3, and GM-CSF, thereby regulating transcription.
- During cytotoxic (CD8+) T cell development, CBF complexes act as transcriptional silencers by binding to the RUNX-binding sequence within the ZBTB7B locus, repressing its expression to allow differentiation.
- CBFB is targeted by HIV-1 Vif protein, which recruits it to form a Cullin-5-RING E3 ubiquitin-protein ligase complex (ECS complex) that catalyzes the ubiquitination and degradation of APOBEC3F and APOBEC3G (PubMed:22190037, PubMed:31792451, PubMed:36598981). The complex can also ubiquitinate APOBEC3H to some extent (PubMed:37640699). Vif association also inhibits the coactivator activity of CBFB (PubMed:22190037).
