Biological Background: CBFB Function and Localization

  • Encoded by the CBFB gene, core-binding factor subunit beta is a transcription factor subunit that forms a heterodimeric complex with RUNX family proteins (RUNX1, RUNX2, RUNX3) known as core-binding factor (CBF). CBFB allosterically enhances the DNA-binding affinity of RUNX transcription factors.
  • CBF complexes bind to the core consensus sequence 5′-TGTGGT-3′ in enhancers and promoters of various genes, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, LCK, IL3, and GM-CSF, thereby regulating transcription.
  • During cytotoxic (CD8+) T cell development, CBF complexes act as transcriptional silencers by binding to the RUNX-binding sequence within the ZBTB7B locus, repressing its expression to allow differentiation.
  • CBFB is targeted by HIV-1 Vif protein, which recruits it to form a Cullin-5-RING E3 ubiquitin-protein ligase complex (ECS complex) that catalyzes the ubiquitination and degradation of APOBEC3F and APOBEC3G (PubMed:22190037, PubMed:31792451, PubMed:36598981). The complex can also ubiquitinate APOBEC3H to some extent (PubMed:37640699). Vif association also inhibits the coactivator activity of CBFB (PubMed:22190037). Related references: PMID:22190037 PMID:31792451 PMID:36598981 PMID:37640699
  • Subcellular localization is predominantly nuclear, consistent with its role in transcriptional regulation.
  • Keywords from UniProt highlight its involvement in alternative splicing, chromosomal rearrangements (e.g., inv(16) in acute myeloid leukemia), proto-oncogenic potential, and disease associations including dwarfism. It is also a phosphoprotein.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein covering amino acids 1-182 of human CBFB. This N-terminal portion contains the conserved domain necessary for RUNX interaction and is identical or highly similar in mouse and rat, supporting the observed cross-reactivity.
  • For western blot (WB), a band near 22 kDa is expected. Consider using nuclear extracts and appropriate loading controls.
  • Immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P) should reveal nuclear localization. Validate in known CBFB-expressing cell lines or tissues.
  • ELISA can be used for quantitative analysis; optimal coating conditions and antibody concentrations should be determined empirically.
  • As with any antibody, validate in the specific experimental context and consider pre-adsorption controls if needed.
