Biological Background: CBFA2T2 Function and Localization
- Protein CBFA2T2 (EHT/MTGR1/ZMYND3/p85) acts as a transcriptional corepressor by recruiting histone‑modifying enzymes and other corepressors to DNA‑binding transcription factors. Related references: PMID:12559562 PMID:15203199
- Through interaction with PRDM14, CBFA2T2 stabilizes PRDM14 and OCT4 on chromatin, playing a key role in embryonic stem cell (ESC) pluripotency and primordial germ cell (PGC) formation. Related references: PMID:27281218
- In a ZBTB33‑dependent manner, CBFA2T2 represses the expression of MMP7. Related references: PMID:23251453
- In acute myeloid leukemia, CBFA2T2 may function in a complex with the RUNX1‑CBFA2T1 fusion protein, contributing to the repression of AML1‑dependent transcription and induction of G‑CSF/CSF3‑dependent cell growth.
- Through heteromerization with CBFA2T3/MTG16, it helps regulate erythroid progenitor proliferation and differentiation by repressing TAL1 target genes.
- CBFA2T2 is required for maintaining the secretory cell lineage in the small intestine; it can inhibit Notch signaling via association with RBPJ and may participate in GFI1‑mediated Paneth cell differentiation.
- The protein contains coiled‑coil and zinc‑finger domains, undergoes phosphorylation and ubiquitination, and is localized exclusively in the nucleus.
- Widely expressed in fetal and adult tissues, with notably high levels in brain, heart, lung, kidney, lymphoid organs, and reproductive tissues.
Experimental Guidance and Technical Tips
- Use this antibody for Western blot detection of endogenous CBFA2T2; expect a band near 67 kDa. Validate the specificity in your sample by using appropriate controls.
- For immunofluorescence/immunocytochemistry (IF/ICC), CBFA2T2 localizes to the nucleus; optimize fixation and permeabilization conditions for your cell type.
- This antibody is suitable for ELISA; the recombinant immunogen (amino acids 325–604) can serve as a positive control or coating antigen.
- When blotting or staining tissues, note that CBFA2T2 is ubiquitously expressed but highly abundant in brain, heart, lung, kidney, and lymphoid tissues; mouse and rat models are appropriate cross-reactive species.
