Biological Background: Caveolin-3 Function and Localization

  • Caveolin-3, also known as M-caveolin, is a scaffolding protein predominantly expressed in muscle tissues, where it localizes to caveolae — small invaginations of the plasma membrane.
  • It functions as a molecular scaffold within caveolar membranes, directly interacting with G-protein alpha subunits and modulating their activity, as well as regulating voltage-gated potassium channels (By similarity).
  • Caveolin-3 plays a critical role in sarcolemma repair in both skeletal and cardiac muscle, facilitating rapid membrane resealing after mechanical stress-induced disruption (By similarity).
  • It mediates the recruitment of CAVIN2 and CAVIN3 proteins to caveolae, essential for caveola formation and stability. Related references: PMID:19262564
  • The protein is localized to the Golgi apparatus membrane and cell membrane, particularly enriched in caveolae and the sarcolemma.
  • Mutations in CAV3 are associated with a spectrum of muscle disorders, including limb-girdle muscular dystrophy type 1C (LGMD1C), rippling muscle disease (RMD2), and long QT syndrome type 9 (LQT9), as well as cardiomyopathy.
  • Post-translational modifications include isopeptide bond formation and ubiquitin conjugation, which may regulate its stability and function.

Experimental Guidance and Technical Tips

  • The immunogen encompasses the full-length human Caveolin-3 (residues 1-151), which should generate antibodies recognizing native and denatured forms; confirm reactivity in the intended sample system.
  • For Western blotting, a band at approximately 17 kDa is expected in human and mouse muscle lysates; use appropriate loading controls and consider protein extraction conditions to preserve membrane proteins.
  • For immunohistochemistry (paraffin), antigen retrieval and detection conditions may need optimization for different muscle tissue types; include positive and negative controls.
  • As a polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative experiments; validated blocking peptides or recombinant protein can be used to confirm specificity.
  • The antibody is also suitable for ELISA; assay conditions should be standardized with recombinant Caveolin-3 protein as a reference.
