Biological Background: Cation-independent M6PR (IGF2R) Function and Localization

  • Also known as 300 kDa mannose-6-phosphate receptor (MPR300), insulin-like growth factor 2 receptor (IGF2R), and CD222, the cation-independent M6PR is a large transmembrane glycoprotein encoded by the IGF2R gene in humans.
  • Functions primarily as a trafficking receptor that binds mannose-6-phosphate (M6P) tags on newly synthesized lysosomal hydrolases in the Golgi, delivering them to endosomes and lysosomes for proper sorting and acidification. Related references: PMID:18817523, PMID:2963003
  • Serves a dual role by also binding insulin-like growth factor 2 (IGF2) at the cell surface, mediating its internalization and clearance, thereby regulating IGF2 availability and signaling. Related references: PMID:18046459
  • Localizes primarily to the Golgi apparatus membrane and endosome membrane, cycling between these compartments through retromer-mediated recycling.
  • Acts as a positive regulator of T-cell coactivation by binding dipeptidyl peptidase 4 (DPP4). Related references: PMID:10900005
  • Post-translational modifications include heavy glycosylation, disulfide bond formation, palmitoylation, acetylation, phosphorylation, and methylation, which are critical for its stability and trafficking functions.
  • Belongs to the mannose-6-phosphate receptor family; the cation-independent form is larger and more multifunctional compared to the cation-dependent M6PR.

Experimental Guidance and Technical Tips

  • Use the immunogen sequence (aa 2140–2300, extracellular domain) to verify specificity, particularly for cell surface binding assays.
  • For western blotting, expect a band around 274 kDa; high glycosylation may cause the protein to migrate at a higher apparent molecular weight. Use 6–8% SDS-PAGE gels and extended transfer times.
  • In immunofluorescence and immunohistochemistry, optimize antigen retrieval and permeabilization to detect both cell surface and Golgi/endosomal pools. Co-localization markers (e.g., GM130 for Golgi, EEA1 for early endosomes) can confirm subcellular specificity.
  • The rabbit PolymAb® format offers high affinity, batch-to-batch consistency, and suitability for multiplex labeling.
  • Validate the antibody in the relevant sample system (e.g., HeLa, HEK293, or tissue sections) to confirm cross-reactivity with mouse and rat targets.
