Biological Background: Catenin alpha-2 Function and Localization
- Catenin alpha-2 (alpha N-catenin, CTNNA2) is a member of the catenin family and functions as a linker between cadherin adhesion receptors and the actin cytoskeleton, playing a critical role in cell-cell adhesion and differentiation in the nervous system (By similarity).
- It is required for proper regulation of cortical neuronal migration and neurite growth, and acts as a negative regulator of the Arp2/3 complex to suppress excessive actin branching (PubMed:30013181). Related references: PMID:30013181
- CTNNA2 regulates morphological plasticity of synapses and contributes to cerebellar and hippocampal lamination during development (By similarity).
- Subcellularly, catenin alpha-2 localizes to the cell membrane, cytoplasm, cytoskeleton, adherens junctions, axon, and nucleus, reflecting its dynamic role in cell adhesion and signaling.
- The protein is predominantly expressed in neural tissues, with strongest expression in fetal and adult brain, including the developing cortical plate and marginal zone of the 20-week-old human fetal brain.
- Key post-translational modifications include phosphorylation, and the protein is subject to alternative splicing. It is associated with disease variants linked to lissencephaly.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 167-290 of human CTNNA2 (isoform 1), a region conserved across human, mouse, and rat, supporting the observed cross-reactivity. Validate the antibody in your specific sample system for optimal performance.
- For Western blot (WB), consider using brain tissue lysates (human, mouse, or rat) as a positive control. A band at approximately 105 kDa is expected; alternative splicing may produce additional isoforms.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval methods and permeabilization steps may be required to access intracellular epitopes due to CTNNA2's multiple subcellular localizations.
- For ELISA applications, the antibody can be used for capture or detection; consider pairing with a matched detection antibody or performing checkerboard titration to optimize the assay.
