Biological Background: CASQ2 Function and Localization

  • CASQ2 encodes calsequestrin-2, also known as the cardiac muscle isoform of calsequestrin (PDIB2), a high-capacity calcium-binding protein located in the sarcoplasmic reticulum lumen.
  • Acts as an internal calcium store in muscle, binding approximately 60 Ca²⁺ ions per molecule via clusters of acidic residues on the protein surface, particularly at subunit interfaces.
  • Regulates the release of lumenal Ca²⁺ through the cardiac ryanodine receptor (RYR2), thereby playing a critical role in excitation-contraction coupling and triggering muscle contraction.
  • Essential for normal cardiac function, contributing to the regulation of heart rate and rhythm.
  • Undergoes post-translational modifications including glycosylation and phosphorylation (UniProt keywords: Glycoprotein, Phosphoprotein).
  • Alternatively spliced transcript variants exist, and mutations in CASQ2 are associated with disease variants (UniProt keywords: Alternative splicing, Disease variant).
  • The protein has a molecular weight of approximately 46.4 kDa (calculated from sequence), consistent with the observed 46 kDa band.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 20-399 of human CASQ2, corresponding to the mature protein after signal peptide cleavage; this design enhances the likelihood of recognizing the native protein.
  • For Western blot (WB), consider using a positive control such as human cardiac tissue lysate and verify a specific band around 46 kDa; optimize antibody dilution and incubation conditions for your specific sample.
  • For immunohistochemistry on paraffin sections (IHC-P), ensure appropriate antigen retrieval and include proper controls to distinguish specific staining.
  • For ELISA applications, titrate the antibody to determine the optimal coating or detection concentration; the antibody can be used in combination with a matched pair antibody if available.
  • Cross-reactivity with mouse and rat has been reported; validate the antibody in these species under your experimental conditions, as reactivity may vary with sample type and protocol.
