Biological Background: Caspase-9 Function and Localization

  • Caspase-9, also known as apoptotic protease Mch-6 (MCH6), APAF-3, or ICE-LAP6, is a member of the cysteine-aspartic acid protease (caspase) family and is encoded by the CASP9 gene in humans.
  • It functions as an initiator caspase in the intrinsic apoptosis pathway. Upon cytochrome c release and Apaf-1 apoptosome formation, caspase-9 is recruited and activated, subsequently cleaving and activating effector caspases-3 and -7. It also cleaves poly(ADP-ribose) polymerase (PARP) and promotes DNA damage-induced apoptosis in an ABL1-dependent manner.   • Alternative splicing of CASP9 produces a catalytically inactive isoform that acts as a dominant-negative inhibitor of caspase-9 signaling.
  • Caspase-9 is expressed ubiquitously, with highest levels in the heart where it is specifically found in myocytes. Moderate expression is observed in liver, skeletal muscle, and pancreas.

  • The antibody is recommended for Western blot (WB), immunofluorescence/immunocytochemistry (IF/ICC), immunohistochemistry (IHC-P), and ELISA. Researchers should optimize dilutions and include appropriate positive and negative controls, especially when working across human, mouse, and rat samples.
