Biological Background: Caspase-8 Function and Localization
- CASP8 encodes a thiol protease that serves as a molecular switch for apoptosis, necroptosis, and pyroptosis, and is essential for preventing tissue damage during embryonic development and adulthood. Related references: PMID:23516580, PMID:35338844, PMID:35446120
- In extrinsic apoptosis, CASP8 is recruited to death receptors (FAS/TNFRSF1A) via the adaptor FADD, forming the death-inducing signaling complex (DISC) where it undergoes proteolytic activation. Related references: PMID:8681376, PMID:8681377, PMID:9184224
