Biological Background: Caspase-8 Function and Localization

- It acts as the initiator caspase in the extrinsic apoptosis pathway, recruited to death receptors (FAS/CD95, TNFRSF1A) via FADD to form the death-inducing signaling complex (DISC), where it is proteolytically activated and subsequently cleaves downstream effector caspases (CASP3, CASP6, CASP7) and BID to trigger mitochondrial cytochrome C release. - For Western blot, a band around 55 kDa is expected, but smaller cleavage fragments may appear upon apoptosis induction. Validate specificity using appropriate positive and negative controls in your cell system.
