Biological Background: Caspase-6 Function and Localization
- Caspase-6 (MCH2) is a cysteine protease belonging to the caspase family, acting as a non-canonical executioner caspase in apoptosis.
- It localizes to the nucleus during apoptosis and cleaves nuclear structural proteins NUMA1 and lamin A/C to induce nuclear shrinkage and fragmentation. Related references: PMID:11953316, PMID:17401638, PMID:8663580
- Under conditions of reduced AMPK phosphorylation, it cleaves BID to trigger cytochrome c release, contributing to hepatocyte apoptosis and nonalcoholic steatohepatitis. Related references: PMID:32029622
- Cleaves key transcription factors such as NF-kappa-B and CREBBP, modulating inflammatory and survival pathways. Related references: PMID:10559921, PMID:14657026
- Serves as a scaffold in ZBP1-mediated PANoptosis, a programmed cell death mechanism integrating pyroptosis, apoptosis, and necroptosis, independently of its proteolytic activity. Related references: PMID:32298652
- Plays an essential role in axon degeneration and pruning during neurogenesis.
- Cleaves the nucleocapsid (N) protein of coronaviruses, including SARS-CoV-2, to regulate viral replication and host interferon responses. Related references: PMID:18155731, PMID:35922005
- Predominantly found in the cytoplasm and nucleus.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 45-162 of human Caspase-6, a region that may help detect both pro- and cleaved forms.
- For WB, expect bands around 33 kDa (proform) and potential cleaved fragments; validate in relevant cell lines or tissues undergoing apoptosis.
- For IF/ICC, expect nuclear and cytoplasmic staining; use appropriate permeabilization methods and validate in your sample system.
- For ELISA, consider pairing with a detection antibody recognizing a different epitope; the antibody can capture or detect target.
