Biological Background: Caspase-4 Function and Localization
- Caspase-4 (also known as TX, Mih1, ICH-2) is an inflammatory caspase and a member of the cysteine-aspartic acid protease (caspase) family.
- It serves as the effector of the non-canonical inflammasome, directly binding lipopolysaccharide (LPS) to induce pyroptosis.

- The immunogen is a synthetic peptide (a.a. 81-270 of human Caspase-4); thus, the antibody may recognize both full-length and cleaved fragments. For WB, consider using reducing conditions and probing around 43 kDa.
- For IHC-P, antigen retrieval conditions may need optimization; validate in human tissue sections known to express caspase-4, such as colonic epithelium.
- ELISA performance can be assessed by direct coating of peptide or recombinant protein; include appropriate controls for specificity.
