Biological Background: Caspase-3 Function and Localization
- Caspase-3 (CPP32, SCA-1) is a thiol protease and the primary executioner caspase in apoptosis. Related references: PMID:18723680, PMID:20566630, PMID:23650375
- Activated by initiator caspases (CASP8, CASP9, CASP10), it then cleaves key substrates like PARP1 (PubMed:10497198, PubMed:16374543, PubMed:7596430), leading to DNA repair inhibition and apoptotic cell death. Related references: PMID:10497198, PMID:16374543, PMID:7596430
- It also cleaves and activates downstream executioner caspases (CASP6, CASP7) and amplifies the caspase cascade (PubMed:7596430). Related references: PMID:7596430
- In addition to apoptosis, Caspase-3 participates in pyroptosis by cleaving gasdermin-E (GSDME) (PubMed:35338844, PubMed:35446120) and regulates inflammation by inactivating interleukin-18 (PubMed:37993714, PubMed:9334240). Related references: PMID:35338844, PMID:35446120, PMID:37993714, PMID:9334240
- It mediates phosphatidylserine exposure on apoptotic cells through cleavage of XKR scramblases (PubMed:23845944, PubMed:33725486) and contributes to cell adhesion changes by cleaving desmosomal proteins (PubMed:17559062). Related references: PMID:23845944, PMID:33725486, PMID:17559062
- Subcellularly, Caspase-3 is predominantly cytoplasmic, consistent with its executioner role.
- Post-translational modifications include acetylation, phosphorylation, S-nitrosylation, and ubiquitination, and it is synthesized as an inactive zymogen that requires cleavage for activation.
- Highly expressed in lung, spleen, heart, liver, kidney, and immune cells, with low expression in testis, reflecting its key role in tissue homeostasis and immune function.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 6-105) of full-length human Caspase-3 (32 kDa). In WB, you may detect both the pro‑form (~32 kDa) and the cleaved large fragment (~17 kDa) depending on apoptosis induction.
- For IHC-P, consider optimizing antigen retrieval conditions (e.g., citrate buffer pH 6.0) as recommended for your tissue type and fixation protocol.
- This polyclonal antibody is reported to cross‑react with mouse and rat Caspase-3 based on sequence homology; however, we recommend validating specificity in your own experimental system.
- For ELISA, use a standard sandwich format and confirm optimal antibody pairing and detection sensitivity with recombinant Caspase-3 as a reference.
