Caspase-3 (CPP32, Apopain, Yama, SCA-1) is a major effector caspase that executes apoptosis by cleaving multiple cellular substrates following activation by initiator caspases.

Experimental Guidance and Technical Tips
- For Western blot, based on the immunogen design (amino acids 176-277, corresponding to the mature large subunit region), the antibody may detect both the 32kDa proenzyme and the 17kDa cleaved fragment. Consider using apoptosis-induced lysates as a positive control.
- For immunohistochemistry on paraffin-embedded tissues, optimal antigen retrieval conditions and antibody dilution should be determined empirically; endogenous peroxidase blocking is recommended.
- The antibody has shown cross-reactivity with mouse and rat Caspase-3, making it suitable for multi-species apoptosis studies. Validate in your specific sample system to confirm reactivity.
