Biological Background: Caspase-3 Function and Localization

  • Caspase-3 (EC 3.4.22.56), also known as apopain, CPP32, Yama, and SCA-1, is a major executioner caspase in the apoptotic pathway. Related references: PMID:18723680, PMID:20566630, PMID:23650375
  • The enzyme is synthesized as an inactive zymogen (pro-caspase-3) that undergoes proteolytic processing by initiator caspases (CASP8, CASP9, CASP10) to generate active p17 and p12 subunits. Related references: PMID:7596430, PMID:18723680
  • Upon activation, Caspase-3 cleaves numerous cellular substrates including PARP1, SREBPs, and caspase-6/-7/-9 to execute apoptosis. Related references: PMID:10497198, PMID:16374543, PMID:7596430
  • Beyond apoptosis, Caspase-3 also mediates pyroptosis by cleaving gasdermin-E (GSDME) and inhibits type I interferon production during viral infection by targeting CGAS, IRF3, and MAVS. Related references: PMID:35338844, PMID:35446120, PMID:30878284
  • The protein localizes primarily to the cytoplasm and is highly expressed in lung, spleen, heart, liver, kidney, and immune cells.
  • Post-translational modifications include acetylation, phosphorylation, S-nitrosylation, and ubiquitination, which regulate its activity and stability.
  • Caspase-3 also plays roles in axon pruning, cell adhesion through RET and DSG2 cleavage, and phosphatidylserine exposure via scramblase cleavage. Related references: PMID:21357690, PMID:17559062, PMID:23845944, PMID:33725486

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 29–175, which covers the p17 large subunit of Caspase-3. In Western blotting, the antibody may recognize both pro-caspase-3 (approx. 32 kDa) and the cleaved active large subunit (approx. 17–20 kDa); appropriate apoptosis induction controls (e.g., staurosporine treatment) are recommended to verify detection of processed forms.
  • The KO validation ensures high target specificity; consider incorporating knockout cell lines or tissues as negative controls in your experiments.
  • For ELISA, validate the antibody pair under the intended assay conditions and confirm sensitivity with recombinant Caspase-3 protein.
  • Cross-reactivity with mouse allows comparative studies between human and mouse apoptosis models.
