Biological Background: Caspase-1 Function and Localization

  • Caspase-1 is a thiol protease that plays a central role in inflammatory responses by proteolytically cleaving the precursors of interleukin-1 beta (IL1B) and interleukin 18 (IL18) into their active mature forms. Related references: PMID:15326478 PMID:15498465 PMID:1574116

  • For Western blot (WB), a band around 45 kDa is expected for the full-length pro-caspase-1; additional lower bands may correspond to processed subunits.
  • For immunofluorescence (IF-P), optimize fixation and permeabilization conditions to preserve both cytoplasmic and membrane-associated signals.
  • Due to the polyclonal nature, lot-to-lot variability may occur; consistently validate each new lot for your specific application.
  • Cross-reactivity with mouse and rat has been reported, but confirm sequence homology in the immunogen region to ensure comparable reactivity across species.
