Biological Background: Caspase-1 Function and Localization
- Caspase-1, also known as Interleukin-1 beta convertase (ICE) or IL-1BC, is a thiol protease that plays a critical initiator role in inflammatory responses and pyroptosis.
- It proteolytically cleaves pro-IL-1β and pro-IL-18 into their active mature cytokines, driving inflammation (PubMed:21147462). It also cleaves Gasdermin-D (GSDMD) to trigger pyroptotic cell death (PubMed:32109412). Related references: PMID:21147462 , PMID:32109412
- Belongs to the caspase family of cysteine proteases; exists as a zymogen that requires processing and assembly into active inflammasome complexes.
- Subcellularly located in the cytoplasm and at the cell membrane.
- Highly expressed in spleen and lung, with lower levels in brain, heart, liver, kidney, testis, and skeletal muscle.
- In response to bacterial infection, caspase-1 also cleaves and activates CASP7, contributing to plasma membrane repair (PubMed:18667412, PubMed:22464733, PubMed:35705808). Related references: PMID:18667412 , PMID:22464733 , PMID:35705808
- During DNA virus infection, it inactivates the cytosolic DNA sensor cGAS by cleavage, modulating antiviral immunity (PubMed:28314590). Related references: PMID:28314590
- Regulated by post-translational modifications including methylation, phosphorylation, and ubiquitin conjugation.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to amino acids 119-296 of mouse CASP1, a region that may be well-conserved and accessible; consider its suitability for detecting both pro-form and cleaved active subunits in your specific assay.
- For Western blot, the predicted molecular weight is 46 kDa for the full-length pro-caspase-1; validate the antibody’s ability to detect cleaved fragments by including appropriate positive controls (e.g., inflammasome-activated lysates).
- ELISA application is reported; perform titration and validation in the mouse sample matrices of interest, and ensure specific detection of caspase-1 over other caspase family members.
