Biological Background: Caspase-1 Function and Localization
- Caspase-1, also known as interleukin-1 beta convertase (ICE) or p45, is a thiol protease and a key member of the cysteine-aspartic acid protease (caspase) family.

- The immunogen corresponds to a region within the p20 subunit of human pro-caspase-1, suggesting the antibody can recognize both the full-length 45 kDa proenzyme and its processed fragments. In Western blot, consider probing lysates from cells treated with inflammasome activators (e.g., LPS/ATP) to observe caspase-1 activation.
- For immunofluorescence (IF-P), validate the antibody in cell lines or tissues with known CASP1 expression (e.g., THP-1 macrophages, spleen), and optimize permeabilization to detect cytoplasmic and membrane-localized pools.
- In ELISA, use the antibody as a capture or detection reagent for caspase-1 quantification; confirm reactivity against recombinant human CASP1 and cross-reactivity with mouse/rat homologs in the relevant sample matrix.
