Biological Background: Caspase-1 Function and Localization
- Caspase-1, also known as interleukin-1 beta convertase (ICE/p45/IL1BC), is a member of the caspase family of cysteine-aspartic acid proteases (thiol proteases) that plays a central role in inflammatory responses and programmed cell death.
- It is essential for the processing and secretion of the pro-inflammatory cytokines interleukin-1 beta (IL-1β) and interleukin-18 (IL-18) by cleaving their precursor forms into biologically active mature peptides.
Related references: PMID:15326478, PMID:15498465, PMID:1574116 - Caspase-1 initiates pyroptosis, a lytic form of programmed cell death, through cleavage of gasdermin-D (GSDMD), with substrate recognition involving an exosite interface distinct from the canonical cleavage site.
Related references: PMID:26375003, PMID:32051255, PMID:32109412, PMID:32553275 - Beyond IL-1β/IL-18 and GSDMD, Caspase-1 also cleaves other substrates including caspase-7 (CASP7) during bacterial infection to promote membrane repair, cyclic GMP-AMP synthase (CGAS) to regulate antiviral immunity, and sphingosine kinase 2 (SPHK2) in apoptotic cells.
Related references: PMID:22464733, PMID:28314590, PMID:20197547 - It is primarily localized in the cytoplasm but can also associate with the cell membrane.
- Tissue expression is highest in spleen and lung, with lower levels detected in liver, heart, small intestine, colon, thymus, prostate, skeletal muscle, peripheral blood leukocytes, kidney, and testis; it is not expressed in the brain.
- Alternative splicing generates isoforms that are inactive in apoptosis, and the protein is subject to post-translational modifications including phosphorylation, ubiquitin-like conjugation, and processing from a zymogen to active enzyme.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 120–297 of human Caspase-1 (NP_150634.1), which lies within the p20 subunit of the processed enzyme; this region may yield antibodies that recognize both the proform and cleaved fragments, but users should validate in their specific sample matrix.
- For Western blotting, the full-length Caspase-1 proenzyme migrates at approximately 45 kDa; additional bands corresponding to processed p20 and p10 subunits may appear depending on activation state—optimize loading and detection conditions accordingly.
- In ELISA applications, consider using the recombinant immunogen as a reference standard or coating antigen, and verify cross-reactivity with related caspase family members if specificity is critical.
- Cross-reactivity has been confirmed for human; for experiments involving other species, perform independent validation as sequence homology varies.
