Biological Background: CASP8AP2 Function and Localization
- CASP8AP2, also known as FLASH, CED-4, or RIP25, is a large scaffold protein encoded by the CASP8AP2 gene, with a predicted molecular weight of approximately 223 kDa.
- It functions as a negative regulator of glucocorticoid receptor (GR) transactivation by suppressing the coactivators NCOA2 and NCOA3 in response to TNF signaling, modulating inflammatory and stress responses. Related references: PMID:12477726, PMID:15698540
- CASP8AP2 is essential for CASP8 activation in FAS-mediated apoptosis and also mediates CASP8-induced NF-κB activation, linking cell death and inflammatory signaling. Related references: PMID:17245429
- It plays an essential role in histone pre-mRNA 3′ end processing, being indispensable for mature histone mRNA production and thereby for proper cell cycle progression through S phase. Related references: PMID:17003125, PMID:19854135, PMID:32367068
- Subcellularly, CASP8AP2 localizes to the nucleus, cytoplasm, PML bodies, and mitochondria, reflecting its multifaceted roles in transcription, apoptosis, and cell cycle control.
- Proteomics data indicate acetylation and phosphorylation of CASP8AP2, suggesting regulatory post-translational modifications that may influence its function in signal transduction.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 1733-1982) of human CASP8AP2. When designing confirmatory experiments, consider using full-length protein or fragments covering this region as positive controls.
- For Western blotting, a reducing SDS-PAGE gel with a high molecular weight marker (approx. 223 kDa) is recommended. Optimize transfer conditions for large proteins.
- In ELISA, titrate the antibody against the immunogen or native CASP8AP2 from cell lysates to determine optimal working dilutions; include appropriate negative controls.
- As a polyclonal antibody, it may recognize multiple epitopes; confirm specificity via knockdown or overexpression systems in the relevant cell line.
- This antibody is reported to react with human CASP8AP2; for other species, verify target sequence homology before use.
