Biological Background: Casein Kinase 2 beta Function and Localization
- Casein Kinase 2 beta (CSNK2B) is the regulatory subunit of the casein kinase II (CK2) complex, regulating the basal catalytic activity of the alpha subunit, a constitutively active serine/threonine-protein kinase that phosphorylates numerous substrates with acidic residues downstream of the target serine/threonine. Related references: PMID:11239457 PMID:16818610
- It forms a tetrameric complex with two catalytic alpha and two regulatory beta subunits, enhancing kinase stability and substrate specificity; the beta subunit is essential for full enzymatic activity.
- CSNK2B participates in the Wnt signaling pathway, contributing to beta-catenin regulation and downstream gene transcription.
- Upon Epstein-Barr virus (EBV) infection, CSNK2B interacts with viral EBNA1, increasing CK2-mediated phosphorylation of PML proteins, which triggers PML polyubiquitylation and degradation, facilitating viral persistence. Related references: PMID:20719947 PMID:24216761
- CSNK2B also suppresses EBV reactivation by mediating ARK2N and JUN at the Z promoter, thereby inhibiting BZLF1 transcription. Related references: PMID:31341047
- The protein localizes predominantly to the nucleus, consistent with its role in phosphorylating nuclear substrates and regulating transcription.
- Post-translational modifications include phosphorylation, acetylation, and ubiquitination; it contains a zinc-binding site. Mutations in CSNK2B are associated with epilepsy and intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen covers the full-length human CSNK2B protein; the antibody may recognize both unmodified and post-translationally modified forms. For phosphorylation-specific analysis, phosphatase treatment may be considered.
- In Western blotting, a band around 25 kDa is expected in lysates from human, mouse, and rat cells; pre-validated with these species but individual sample optimization is recommended.
- For immunohistochemistry (paraffin sections), nuclear staining is anticipated; antigen retrieval conditions and primary antibody dilution should be optimized for the specific tissue.
- For ELISA, the recombinant immunogen can serve as a positive control; validate assay conditions using the relevant sample matrix.
