Biological Background: Troponin I, cardiac muscle Function and Localization

  • Troponin I (TnI) is the inhibitory subunit of the troponin complex, which regulates striated muscle contraction by conferring calcium sensitivity to the actomyosin ATPase activity.
  • Cardiac troponin I (cTnI) is specifically expressed in cardiac muscle and is a well-established biomarker for myocardial injury.
  • The TNNI3 gene encodes the cardiac isoform, and mutations in this gene are linked to inherited cardiomyopathies such as hypertrophic and restrictive cardiomyopathy.
  • As an actin-binding protein, cTnI binds to actin-tropomyosin in the absence of calcium, inhibiting contraction; calcium binding to troponin C relieves this inhibition.
  • Post-translational modifications, including phosphorylation and acetylation, modulate its function and are critical in cardiac physiology and disease.
  • The protein shares conserved domains with other troponin I isoforms (slow-twitch and fast-twitch skeletal muscle), but the N-terminal extension is unique to the cardiac form and contains several phosphorylation sites.
  • cTnI localizes to the sarcomeric thin filament, contributing to the structural integrity and calcium-dependent regulation of cardiac myofibrils.

Experimental Guidance and Technical Tips

  • The immunogen covers the full-length human cTnI (aa 1-210), suggesting the antibody recognizes the majority of the protein sequence. When used in Western blotting, expect a band around 24 kDa in human, mouse, and rat cardiac tissue lysates.
  • For immunoprecipitation (IP), consider using 1–2 µg of antibody per 500 µg of protein lysate and optimize based on the target abundance. Include appropriate controls to confirm specificity.
  • In immunofluorescence paraffin-embedded sections (IF-P), a heat-induced antigen retrieval step in citrate buffer (pH 6.0) is often beneficial for exposing epitopes. Test with cardiac muscle tissue sections.
  • For ELISA, this antibody may be used as a capture or detection reagent. Pair it with a monoclonal anti-cTnI to develop a sensitive sandwich assay; cross-reactivity with mouse and rat cTnI can be advantageous for preclinical studies.
  • Since the antibody cross-reacts with mouse and rat, it is suitable for translational research using common animal models. However, always validate in the relevant species and sample matrix.
