Biological Background: CARD8 Function and Localization
- CARD8 (Caspase recruitment domain-containing protein 8), also known as CARDINAL, TUCAN, NDPP1, DAKAR, DACAR, or NDPP, is a member of the CARD-containing protein family.
- CARD8 acts as an inflammasome sensor, recognizing pathogen/damage signals such as HIV-1 protease activity or the Val-boroPro inhibitor, and mediates inflammasome activation leading to pyroptosis of CD4+ T-cells and macrophages.

- Upon pathogen detection, the N-terminal fragment is ubiquitinated and degraded by the proteasome, releasing the C-terminal fragment to form the CARD8 inflammasome, which directly recruits pro-caspase-1 and promotes its activation, resulting in GSDMD cleavage and pyroptosis.
Related references: PMID:32558991, PMID:32051255, PMID:32840892, PMID:33053349, PMID:34019797 - In addition to its inflammasome role, CARD8 also functions as a negative regulator of NF-kappa-B activation and the NLRP3 inflammasome.
Related references: PMID:11551959, PMID:12067710, PMID:24517500 - CARD8 is localized in the cytoplasm, nucleus, and assembled inflammasome complexes.
- CARD8 is highly expressed in lung, ovary, testis, and placenta, with lower expression in heart, kidney, and liver; it is also expressed in spleen, lymph node, and bone marrow.
Related references: PMID:11551959, PMID:11821383 - CARD8 plays a role in host defense against HIV-1: it senses HIV-1 protease activity, leading to clearance of latent virus after viral reactivation, while HIV-1 can evade sensing by maintaining protease inactivity in infected cells.

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 1-200 amino acid region, so the antibody likely recognizes the full-length CARD8 and its N-terminal fragment. Consider verifying specificity by analyzing autoproteolytic processing products in cell lysates.
- For western blot, use human cell lines or tissues with confirmed CARD8 expression (e.g., THP-1 macrophages, lung or ovary lysates). The predicted full-length protein is ~61 kDa, but cleavage fragments may also be detected. Include appropriate positive and negative controls.
- For ELISA, the antibody can be employed in sandwich or indirect formats. Pair with a detector antibody against a distinct CARD8 epitope to ensure assay specificity.
- As this polyclonal antibody is validated for human CARD8, cross-reactivity with other species should be tested experimentally. Consider using inflammasome activators (e.g., Val-boroPro) or HIV-1 protease inhibitors in relevant infection models to study CARD8 dynamics.
