Biological Background: CARD14 Function and Localization
- Acts as a scaffolding protein that activates NF-κB and p38/JNK MAP kinase pathways by forming a complex with BCL10 and MALT1, which is required for MALT1 proteolytic activity and inflammatory gene expression. Related references: PMID:11278692, PMID:21302310, PMID:27071417
- A splice variant may function as a dominant negative regulator of NF-κB, unable to activate the pathway. Related references: PMID:21302310, PMID:26358359
- Contains card, coiled-coil, and MAGUK domains, linking it to apoptosis and inflammatory signaling.
- Localizes predominantly to the cytoplasm.
- Isoform 1 is expressed in placenta and epidermal keratinocytes; isoform 2 is detected in leukocytes and fetal brain.
- Phosphorylated form is indicated; associated with disease variants, particularly psoriasis.
- May also protect cells from apoptosis by participating in TRAF2/3/6-mediated signaling.

- For Western blot, the predicted molecular weight is ~113 kDa; use a gel percentage and transfer conditions appropriate for high molecular weight proteins.
- For immunohistochemistry on paraffin-embedded tissues, antigen retrieval may be required; consider testing on known positive tissues such as placenta or skin.
