- CARD11 (Caspase recruitment domain-containing protein 11, also known as CARMA1) is an adapter protein critical for adaptive immunity, transducing NF-kB activation downstream of T-cell receptor (TCR) and B-cell receptor (BCR) engagement. - Upon activation, CARD11 homooligomerizes to form a nucleating helical template, recruiting BCL10 via CARD-CARD interactions, which then promotes BCL10 polymerization and MALT1 recruitment; this CBM complex triggers I-kappa-B kinase (IKK) phosphorylation, leading to NF-kB nuclear translocation and activation of MAP kinase p38 pathways.
