Biological Background: CAPZA1 Function and Localization
- CAPZA1 encodes F-actin-capping protein subunit alpha-1, also known as CapZ alpha-1, which is a key regulator of actin filament dynamics.
- The protein binds in a Ca2+-independent manner to the barbed ends of actin filaments, blocking subunit exchange without severing filaments.
- CAPZA1 may play a role in the formation of epithelial cell junctions. Related references: PMID:22891260
- Together with the beta subunit (CAPZB), it forms the barbed end of the dynactin complex, stabilizing the complex and activating dynein for transport along microtubules.
- Subcellularly, CAPZA1 is localized to the cytoplasm and cytoskeleton.
- The protein is subject to acetylation and phosphorylation, and it contains structural features determined by X-ray crystallography.
Experimental Guidance and Technical Tips
- As the immunogen covers full-length human CAPZA1 (amino acids 1–286), the antibody is expected to detect endogenous CAPZA1 in whole-cell lysates. For Western blot, consider using a denaturing gel and optimizing antibody dilution in the relevant sample system.
- For ELISA, evaluate direct or indirect detection formats depending on your assay requirements.
- Given its cross-reactivity with human, mouse, and rat, this antibody can be applied across species, but always validate performance in the specific sample context.
- Since CAPZA1 is involved in actin dynamics and cell junctions, the antibody may be useful for studying cytoskeletal reorganization, cell migration, or epithelial integrity.
- The predicted molecular weight is approximately 33 kDa; confirm bands with a molecular weight marker in your experimental setup.
