Biological Background: Caprin-2 Function and Localization
- Caprin-2 (also known as EEG-1, C1QDC1, RNG140) is an RNA-binding protein implicated in the regulation of mRNA transport and translation, particularly in neuronal systems.
- It promotes phosphorylation of the Wnt coreceptor LRP6, leading to increased activity of the canonical Wnt signaling pathway (PubMed:18762581). It also facilitates constitutive LRP6 phosphorylation by CDK14/CCNY during the G2/M phase of the cell cycle, potentially priming cells for Wnt responsiveness (PubMed:27821587).

- Caprin-2 modulates the expression of proteins involved in synaptic plasticity, likely through its role in mRNA translation control (By similarity).
- It is involved in regulating growth as erythroblasts shift from a highly proliferative state toward terminal differentiation, and may also function in apoptosis (PubMed:14593112).
Related references: PMID:14593112
- Subcellular localization includes the cytoplasm, mitochondrion, and cell membrane, suggesting multifaceted cellular roles.
- Tissue expression is broad, with the highest levels detected in the brain and spleen.
- Multiple isoforms are generated by alternative splicing, and the protein features a coiled-coil domain, calcium- and metal-binding properties, and undergoes phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 88–329 of human CAPRIN2, a region that may be conserved across species; cross-reactivity with human and rat is confirmed.
- For western blotting, a band near 126 kDa is expected; however, due to phosphorylation and alternative splicing, additional or shifted bands may appear under certain conditions.
- When using ELISA, consider pairing with a capture antibody targeting a distinct epitope if developing a sandwich assay, and validate the antibody pair in the relevant sample matrix.
- We recommend including appropriate positive and negative controls (e.g., CAPRIN2-overexpressing lysates or siRNA knockdown) and optimizing detection conditions for your specific experimental system.
