Biological Background: CAPRIN1 Function and Localization
-
Acts as an mRNA-binding protein that regulates mRNA transport, translation, and stability. It directly binds to MYC and CCND2 mRNAs, and in neuronal cells, to BDNF, CAMK2A, CREB1, MAP2, NTRK2, GRIN1, and KPNB1 mRNAs. Related references: PMID:17210633
-
Essential for cytoplasmic stress granule formation, mediating phase separation to assemble mRNAs into ribonucleoprotein granules that inhibit translation and promote deadenylation. Related references: PMID:35977029 PMID:31439799 PMID:32302570 PMID:32302571
-
Involved in neurogenesis and synaptic plasticity; regulates cell proliferation and migration in multiple cell types. Related references: PMID:35979925
-
Localizes to the cytoplasm, cytoplasmic ribonucleoprotein granules, dendrites, and lamellipodia.
-
Ubiquitously expressed across tissues.
-
Post-translationally modified by phosphorylation, acetylation, and methylation; is a glycophosphatidylinositol (GPI)-anchored membrane protein. Disease-associated variants are implicated in autism spectrum disorder, intellectual disability, and neurodegeneration.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (amino acids 1-300) of human CAPRIN1. As a polyclonal antibody, it recognizes multiple epitopes, which may enhance detection sensitivity.
- For Western blot (WB), use whole-cell lysates from human, mouse, or rat cell lines as positive controls, given the ubiquitous expression of CAPRIN1. The predicted molecular weight is approximately 78 kDa, though post-translational modifications may cause band shifts.
- In immunofluorescence/ICC, expect cytoplasmic and granular staining patterns. Validate fixation and permeabilization conditions for your cell type.
- For ELISA, the antibody may be used as a capture or detection reagent; determine optimal concentrations empirically.
- This antibody has not been validated for other applications such as IHC or IP; consider in-house testing if needed.
