- CAMKK2 (Calcium/calmodulin-dependent protein kinase kinase 2), also known as CaM-KK beta, is a serine/threonine kinase that belongs to a proposed calcium-triggered signaling cascade.
- It phosphorylates and activates downstream kinases including CAMK1, CAMK4, and AMPK, thereby regulating various cellular processes (By similarity).
- Multiple isoforms exist due to alternative splicing; isoform 1 and 2 phosphorylate CAMK1 and CAMK4, isoform 3 additionally targets CAMK1D, while isoforms 4–6 are inactive due to partial calmodulin-binding domain loss.
- The protein is involved in hippocampal activation of CREB1 and may play roles in neurite growth, with isoform 3 promoting elongation and isoform 1 promoting branching (By similarity).
- Subcellularly, CAMKK2 is found in the nucleus, cytoplasm, and neuronal projections.
- It is ubiquitously expressed with highest levels in the brain, and intermediate levels in spleen, prostate, thyroid and leukocytes; lowest in lung.
