Biological Background: CaMKI Function and Localization
- Calcium/calmodulin-dependent protein kinase type 1 (CaMKI, also known as CaMKI-alpha) is a serine/threonine kinase that operates in the calcium-triggered CaMKK-CaMK1 signaling cascade, regulating diverse processes including transcription, cell cycle progression, hormone production, cell differentiation, actin filament organization, and neurite outgrowth.
- CaMKI phosphorylates a range of substrates, including ARHGEF7/BETAPIX, MARK2, HDAC5, NUMB, CREB1, ATF1, CFTR, MYL9, and SYN1/synapsin I, thereby mediating effects on spine formation, axonal extension, growth cone motility, and long-term potentiation in neurons.
- The protein is widely expressed, with notable presence in the zona glomerulosa of the adrenal cortex, where it plays a role in K(+) and ANG2-mediated regulation of aldosterone synthase (CYP11B2) to produce aldosterone.
- Subcellular localization includes both the cytoplasm and the nucleus, enabling CaMKI to interact with nuclear and cytoplasmic targets; it is involved in the nuclear export of HDAC5 to regulate muscle cell differentiation.
- Keywords associated with CaMKI include 3D-structure, allosteric enzyme, ATP-binding, calmodulin-binding, cell cycle, cytoplasm, developmental protein, differentiation, neurogenesis, phosphoprotein, serine/threonine-protein kinase, and Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen used to raise this antibody corresponds to the C-terminal region (amino acids 271-370) of human CaMKI. Consequently, the antibody may preferentially detect the full-length protein and could also recognize any isoforms containing this epitope.
- For Western blot (WB) analysis, an expected band at approximately 41 kDa has been reported; perform appropriate positive and negative controls to confirm specificity in your sample type.
- In immunofluorescence/immunocytochemistry (IF/ICC), consider that CaMKI localizes to both cytoplasmic and nuclear compartments; optimal fixation and permeabilization conditions may need to be empirically determined.
- For ELISA applications, validate the antibody’s reactivity with your target antigen using a standard curve and ensure the capture/detection format is optimized for the polyclonal nature of the reagent.
- Cross-reactivity has been documented for human and mouse; if using other species, assess the sequence homology of the immunogen region before proceeding.
