- Calcium/calmodulin-dependent serine/threonine-protein kinase that phosphorylates transcription activators such as CREB1, MEF2D, JUN, and RORA, playing key roles in immune response, inflammation, and memory consolidation.
- Operates in the CaMKK-CaMK4 signaling cascade and can activate MAP kinases (MAPK1/ERK2, MAPK8/JNK1, MAPK14/p38), stimulating transcription through ELK1 and ATF2 phosphorylation.
- In the thymus, regulates CD4(+)/CD8(+) double positive thymocyte selection; in CD4 memory T-cells, links TCR signaling to production of IL2, IFNG, and IL4.
- Mediates dendritic cell survival by connecting TLR4 signaling to temporal BCL2 expression, and regulates differentiation and survival of osteoclasts.
- Phosphorylates CREB1 on Ser-133 in hippocampal neuron nuclei, contributing to long-term potentiation (LTP) and memory consolidation.
- Localizes to both the cytoplasm and nucleus.
- Expressed in brain, thymus, CD4 T-cells, testis, and epithelial ovarian cancer tissue.
