Biological Background: CAMK1G Function and Localization
- CAMK1G (Calcium/calmodulin-dependent protein kinase type 1G) is a serine/threonine-protein kinase belonging to the CaM kinase family.
- Also known as CaM kinase I gamma, CaM-KI gamma, CaMKI gamma, CaMKIG, and CLICK III (CaMK-like CREB kinase III).
- Functions in a calcium-triggered signaling cascade; in vitro, it phosphorylates transcription factor CREB1 (By similarity).
- Localizes to the cytoplasm, Golgi apparatus membrane, and cell membrane.
- Predominantly expressed in brain, with high expression in forebrain neocortex, striatum, and limbic system; also detected at lower levels in skeletal muscle, kidney, spleen, and liver.
- Contains conserved ATP-binding, calmodulin-binding, and kinase domains; undergoes prenylation for membrane anchoring.
- Alternative splicing generates multiple isoforms.
- Keywords: ATP-binding, calmodulin-binding, kinase, serine/threonine-protein kinase, prenylation, lipoprotein, membrane, Golgi apparatus, cytoplasm, reference proteome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 217–476), which may detect multiple isoforms; consider validating in your specific sample system.
- For western blot (WB), use appropriate positive controls (e.g., brain tissue lysates) due to enriched expression in the central nervous system.
- ELISA: consider employing purified recombinant CAMK1G protein as a standard; cross-reactivity with mouse and rat suggests utility in rodent model studies.
- Verify antibody reactivity in your species of interest, as cross-reactivity is reported but may vary with sample type or experimental conditions.
