Biological Background: CAMK1D Function and Localization

  • CAMK1D, also known as CaM kinase I delta or CKLiK, is a serine/threonine-protein kinase encoded by the CAMK1D gene and a member of the CaM kinase family.
  • Operates in the calcium-triggered CaMKK-CaMK1 signaling cascade, activating CREB-dependent gene transcription, regulating granulocyte respiratory burst, and promoting hippocampal neuron dendritic growth.
  • Subcellularly, CAMK1D localizes to both cytoplasm and nucleus.
  • Widely expressed across tissues, with highest levels in polymorphonuclear leukocytes (neutrophilic and eosinophilic granulocytes) and low to undetectable in monocytes and lymphocytes.
  • May contribute to apoptosis in erythroleukemia cells and can phosphorylate transcription factor CREM isoform Beta.
  • Features calmodulin-binding and ATP-binding domains, undergoes alternative splicing, and is regulated by allostery; implicated in inflammatory response and neurogenesis.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal 1-180 amino acids of human CAMK1D, a region conserved across human, mouse, and rat, consistent with the reported cross-reactivity. For Western blot (WB), expect a band near 43 kDa. Optimize blocking and incubation steps for each application.
  • For immunofluorescence (IF/ICC), choose fixation/permeabilization protocols that preserve nuclear and cytoplasmic epitopes; verify staining in a cell line with confirmed expression (e.g., HL-60 for granulocytic lineage).
  • As a polyclonal antibody, lot-to-lot performance should be validated. Potential cross-reactivity with other CaMK kinases may occur; specificity can be confirmed using knockout or knockdown controls.
