Biological Background: CAMK1D Function and Localization
- CAMK1D, also known as CaM kinase I delta (CaMKI delta, CaMKID) or CKLiK, is a Ca²⁺/calmodulin-dependent serine/threonine kinase and a member of the CaMK1 family of protein kinases.
- Encoded by the CAMK1D gene, it functions in the calcium-triggered CaMKK-CaMK1 signaling cascade, where it activates CREB-dependent gene transcription upon calcium influx and regulates processes such as granulocyte function and hippocampal neuron dendritic growth.
- In neutrophils, CAMK1D is required for cytokine-induced proliferation and activation of the respiratory burst; it directly phosphorylates transcription factors CREB1 and CREM isoform Beta in vitro.
- The protein localizes to both the cytoplasm and the nucleus, consistent with its roles in signal transduction and transcriptional regulation.
- CAMK1D is widely expressed but shows highest expression in polymorphonuclear leukocytes (neutrophils and eosinophils), with little or no expression in monocytes and lymphocytes.
- Post-translational modifications include phosphorylation, ubiquitination (Ubl conjugation), and allosteric activation by Ca²⁺/calmodulin binding; alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-180) of human CAMK1D, a domain highly conserved across species; the antibody is suitable for WB, IF/ICC, and ELISA.
- In western blot, the predicted molecular weight is 43 kDa, but additional bands may appear due to isoforms or post-translational modifications; consider optimizing blocking and dilution conditions.
- For immunofluorescence/immunocytochemistry, appropriate fixation and permeabilization are recommended to detect cytoplasmic and nuclear CAMK1D signals.
- Cross-reactivity with mouse and rat has been confirmed; however, it is advisable to validate performance in your specific sample system and application.
