- Calumenin (also known as Crocalbin and IEF SSP 9302) is a calcium-binding protein encoded by the CALU gene.
- It plays a regulatory role in vitamin K-dependent carboxylation, where it inhibits gamma-carboxylase GGCX activity on N-terminal glutamate residues.
- The protein binds up to 7 calcium ions with low affinity and is involved in secretory pathway calcium homeostasis.
- Calumenin localizes primarily to the endoplasmic reticulum membrane and Golgi apparatus, and is also found in the sarcoplasmic reticulum lumen, melanosomes, and can be secreted.
- It is ubiquitously expressed, with highest levels in heart, placenta, and skeletal muscle, lower in lung, kidney, and pancreas, and very low in brain and liver.
