Biological Background: Calreticulin Function and Localization
- Calreticulin (also known as CRP55, Calregulin, ERp60, HACBP, grp60) is a highly conserved calcium-binding chaperone predominantly located in the endoplasmic reticulum (ER) lumen.
- It promotes proper protein folding, oligomeric assembly, and quality control of newly synthesized glycoproteins through the calreticulin/calnexin cycle, interacting transiently with monoglucosylated substrates. Related references: PMID:7876246
- Calreticulin interacts with the DNA-binding domain of the glucocorticoid receptor (NR3C1) and facilitates its nuclear export, linking it to transcriptional regulation. Related references: PMID:11149926
- Although primarily an ER luminal protein, calreticulin is also found in the cytosol, on the cell surface, secreted into the extracellular matrix, and within specialized vesicles such as cortical and cytolytic granules.
