Biological Background: Calreticulin Function and Localization
- Calreticulin, also known as CRP55, calregulin, ERp60, HACBP, and grp60, is encoded by the CALR gene.
- It functions as a calcium-binding chaperone that promotes the folding, oligomeric assembly, and quality control of newly synthesized glycoproteins in the endoplasmic reticulum via the calreticulin/calnexin cycle.
- As an ER resident lectin, it interacts transiently with nearly all monoglucosylated glycoproteins synthesized in the ER. Related references: PMID:7876246
- Calreticulin also mediates the nuclear export of the glucocorticoid receptor (NR3C1) by binding to its DNA-binding domain. Related references: PMID:11149926
- Subcellularly, it is primarily found in the ER lumen, but is also present in the cytosol, on the cell surface, and in the extracellular matrix, as well as in cytoplasmic vesicles such as cortical and cytolytic granules.
- The protein undergoes several post-translational modifications, including disulfide bond formation, acetylation, hydroxylation, and N-linked glycosylation, contributing to its structural and functional regulation.

- The immunogen is a recombinant fusion protein spanning amino acids 18–203 of human Calreticulin (NP_004334.1), corresponding to the N-terminal domain; this region is highly conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blot (WB), a specific band at around 47 kDa is expected; use appropriate loading controls and validate in cell or tissue lysates known to express Calreticulin.
- KO validation confirms antibody specificity; consider using isogenic wild-type and Calreticulin knockout cell lines as negative controls in your experiments.
