Biological Background: Calreticulin Function and Localization

  • Calreticulin, also known as CRP55, calregulin, ERp60, HACBP, and grp60, is encoded by the CALR gene.
  • It functions as a calcium-binding chaperone that promotes the folding, oligomeric assembly, and quality control of newly synthesized glycoproteins in the endoplasmic reticulum via the calreticulin/calnexin cycle.
  • As an ER resident lectin, it interacts transiently with nearly all monoglucosylated glycoproteins synthesized in the ER. Related references: PMID:7876246
  • Calreticulin also mediates the nuclear export of the glucocorticoid receptor (NR3C1) by binding to its DNA-binding domain. Related references: PMID:11149926
  • Subcellularly, it is primarily found in the ER lumen, but is also present in the cytosol, on the cell surface, and in the extracellular matrix, as well as in cytoplasmic vesicles such as cortical and cytolytic granules.
