Biological Background: Calreticulin Function and Localization

  • Calreticulin (CALR) is a 417-amino acid calcium-binding protein, also known as CRP55, Calregulin, ERp60, HACBP, and grp60, encoded by the CALR gene in humans.
  • It functions as a lectin chaperone in the endoplasmic reticulum (ER), interacting transiently with monoglucosylated glycoproteins to promote folding, oligomeric assembly, and quality control through the calreticulin/calnexin cycle (PubMed:7876246). Related references: PMID:7876246
  • Calreticulin also mediates the nuclear export of NR3C1 (glucocorticoid receptor) by binding to its DNA-binding domain, influencing transcriptional regulation (PubMed:11149926). Related references: PMID:11149926
  • It is localized primarily in the ER lumen but can also be found in the cytosol, secreted into the extracellular space, associated with the cell surface, and within cytoplasmic vesicles such as secretory granules and lysosomes.
  • Calreticulin plays a role in calcium homeostasis and is involved in maternal gene expression regulation; it may participate in oocyte maturation and the block to polyspermy upon cortical granule exocytosis (By similarity).
  • The protein contains a signal sequence, three calreticulin family repeats, and undergoes post-translational modifications including acetylation, hydroxylation, glycosylation, and disulfide bond formation, with zinc and calcium binding properties.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 18-203, which encompasses the N-terminal domain and part of the P-domain, known to be highly conserved. This design likely yields an antibody recognizing native and denatured forms, suitable for multiple applications.
  • For Western blotting (WB), a primary band at approximately 47kDa is expected; consider using reducing conditions and validate in the relevant cell or tissue lysate. Titrate antibody dilutions to achieve optimal signal-to-noise.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), fixation and permeabilization steps may influence staining of ER-localized calreticulin; antigen retrieval with citrate buffer (pH 6.0) and blocking with serum are commonly used.
  • For ELISA, this monoclonal antibody can serve as a capture or detection reagent; pairing with a matched antibody or calibrator is recommended for quantitative assays.
  • The KO validation status indicates that the antibody specifically recognizes calreticulin, as signal is abolished in knockout models; always include appropriate controls in experimental setups.
