- Calponin-1, also known as basic calponin or calponin H1, is a smooth muscle-specific isoform encoded by the CNN1 gene. Alternative splicing generates additional variants.
- It is a thin filament-associated protein that binds actin, calmodulin, and tropomyosin, thereby regulating smooth muscle contraction by inhibiting actomyosin Mg-ATPase activity.
- Calponin-1 belongs to the calponin family of actin-binding proteins, which share a conserved calponin homology (CH) domain and are involved in cytoskeletal organization.
- Subcellularly, calponin-1 localizes to the contractile apparatus in smooth muscle cells, often associated with actin stress fibers.
- Post-translational modifications, particularly phosphorylation, modulate calponin-1’s interaction with actin and its inhibitory function, linking it to signaling pathways that control muscle tone.
